It should appear immediately on any posts you make after updating your profile with a new sig.
On older posts it will take longer, which relates to how the software updates threads. Normally it will update whenever someone posts to the thread, I don't know how it updates other threads (but, eventually even the threads in Limbo/Oblivion, which no one can post to, update).

I suspect that the quote marks are messing with our software's crude parsing capability. Perhaps try a URL shortener, and use that instead.
